Journal ArticlePublication PTPN11 induces endoplasmic stress and apoptosis in SH-SY5Y cells(Elsevier Ltd, 2017-11-19)PTPN11 is associated with regulation of growth factor signaling pathways in neuronal cells. Using SH-SY5Y neuroblastoma cells, we showed that adeno-associated virus (AAV)-mediated PTPN11 upregulation was associated with TrkB antagonism, reduced neuritogenesis and enhanced endoplasmic reticulum (ER) stress response leading to apoptotic changes. Genetic knock-down of PTPN11 on the other hand leads to increased TrkB phosphorylation in SH-SY5Y cells. ER stress response induced by PTPN11 upregulation was alleviated pharmacologically by a TrkB agonist. Conversely the enhanced ER stress response induced by TrkB receptor antagonism was ameliorated by PTPN11 suppression, providing evidence of cross-talk of PTPN11 effects with TrkB actions. BDNF treatment of neuronal cells with PTPN11 upregulation also resulted in reduced expression of ER stress protein markers. This study provides evidence of molecular interactions between PTPN11 and the TrkB receptor in SH-SY5Y cells. The results reinforce the role played by PTPN11 in regulating neurotrophin protective signaling in neuronal cells and highlight that PTPN11 dysregulation promotes apoptotic activation. Based on these findings we suggest that blocking PTPN11 could have potential beneficial effects to limit the progression of neuronal loss in neurodegenerative disorders.

Publication Open AccessJournal ArticleGlaucoma is associated with plasmin proteolytic activation mediated through oxidative inactivation of neuroserpin(Nature Publishing Group, 2017-08-21)Neuroserpin is a serine protease inhibitor that regulates the activity of plasmin and its activators in the neuronal tissues. This study provides novel evidence of regulatory effect of the neuroserpin on plasmin proteolytic activity in the retina in glaucoma. Human retinal and vitreous tissues from control and glaucoma subjects as well as retinas from experimental glaucoma rats were analysed to establish changes in plasmin and neuroserpin activity. Neuroserpin undergoes oxidative inactivation in glaucoma which leads to augmentation of plasmin activity. Neuroserpin contains several methionine residues in addition to a conserved reactive site methionine and our study revealed enhanced oxidation of Met residues in the serpin under glaucoma conditions. Met oxidation was associated with loss of neuroserpin inhibitory activity and similar findings were observed in the retinas of superoxide dismutase (SOD) mutant mice that have increased oxidative stress. Treatment of purified neuroserpin with H2O2 further established that Met oxidation inversely correlated with its plasmin inhibitory activity. Dysregulation of the plasmin proteolytic system associated with increased degradation of the extracellular matrix (ECM) proteins in the retina. Collectively, these findings delineate a novel molecular basis of plasmin activation in glaucoma and potentially for other neuronal disorders with implications in disease associated ECM remodelling.

Journal ArticlePublication Amyloid β accumulation and inner retinal degenerative changes in Alzheimer’s disease transgenic mouse(Elsevier Ireland Ltd, 2016-06-03)The APP-PS1ΔE9 mouse model of Alzheimer’s disease (AD) exhibits age dependent amyloid β (Aβ) plaque formation in their central nervous system due to high expression of mutated human APP and PSEN1 transgenes. Here we evaluated Aβ deposition and changes in soluble Aβ accumulation in the retinas of aged APP-PS1 mice using a combination of immunofluorescence, retinal flat mounts and western blotting techniques. Aβ accumulation in the retina has previously been shown to be associated with retinal ganglion cell apoptosis in animal models of glaucoma. This study investigated changes in the inner retinal function and structure in APP-PS1 mice using electrophysiology and histological approaches respectively. We report for the first time a significant decline in scotopic threshold response (STR) amplitudes which represents inner retinal function in transgenic animals compared to the wild type counterparts (p < 0.0001). Thinning of the retina particularly involving inner retinal layers and reduction in axonal density in the optic nerve was also observed. TUNEL staining was performed to examine neuronal apoptosis in the inner retina. Intraocular pressure (IOP) measurements showed that APP-PS1ΔE9 mice had a slightly elevated IOP, but the significance of this finding is not yet known. Together, these results substantiate previous observations and highlight that APP-PS1ΔE9 mice show evidence of molecular, functional and morphological degenerative changes in the inner retina.

Identification of various activators and inhibitors of the TrkB receptor and greater understanding their binding mechanisms is critical to elucidate the biochemical and pharmacological pathways and analyse various protein crystallization studies. The data presented here is related to the research article entitled “Brain Derived neurotrophic factor is involved in the regulation of glycogen synthase kinase 3β (GSK3β) signalling” [1]. Cyclotraxin B (CTXB) is a disulphide bridge linked cyclic peptide molecule that interacts with TrkB receptor and inhibits the BDNF/TrkB downstream signalling. This article reports for the first time binding mechanism and interaction parameters of CTXB with the TrkB receptor. The molecular model of CTXB has been generated and it’s docking with TrkB domain carried out to determine the critical residues involved in the protein peptide interaction.

Publication Open AccessJournal ArticleRXR agonist, 9-cis-13,14-dihydroretinoic acid (9CDHRA), reduces damage and protects from demyelination in transsynaptic degeneration model(Elsevier Ltd, 2024-11)Neurodegenerative and demyelinating disease, such as multiple sclerosis (MS) are at the forefront of medical research and the discovery of new drugs and therapeutics. One phenomenon of degeneration seen in these diseases is transsynaptic degeneration (TSD), where damage from one axon spreads to the other axons that are connected to it synaptically. It has previously been found that demyelination occurs prior to neuronal loss in an experimental form of induced TSD. Retinoid-x receptor (RXR) agonists have been shown to promote remyelination. Therefore, this study aimed to reveal the effects of a novel endogenous RXR-γ agonist, 9-cis-13,14-dihydroretinoic acid (9CDHRA), on preventing or restoring the effects of TSD. 9CDHRA was administered to mice following optic nerve crush (ONC) procedures, and electrophysiology (visual evoked potential, VEP) and histological (immunofluorescent) assessments were performed. It was found that 9CDHRA treatment effectively delayed glial activation and reduced the presence of apoptosis at the site of injury and further anterogradely in the visual system, including the lateral geniculate nucleus (LGN) and primary visual cortex (V1). Most notably, 9CDHRA was able to maintain myelin levels following ONC, and effectively protected from demyelination. This was corroborated by VEP recordings with improved P1 latency. The promising findings regarding the injury attenuating and myelin protecting properties of 9CDHRA necessitates further investigations into the potential therapeutic uses of this compound.

Conference PublicationPublication Orgasmic Gushing: where does the fluid come from and how is it produced?(Women in Research (WiR), 2005)There are three sexual fluids from women: lubrication (e.g. transudation of fluid across the mucosa of the vagina, and mucus from the greater vestibular glands); female ejaculation (from paraurethral glands); and gushing. Orthodox western medicine and physiology does not yet have a standardized description or explanation for the third, gushing. The present paper proposes that the gushing fluid is a filtrate of plasma, produced by the mechanism known as transudation. This is an additional application for the transudation mechanism, after the well accepted roles in lubrication of the vagina, and in generating serous fluids. DatasetPublication Mapping Long Term Changes in Mangrove Cover and Predictions of Future Change under Different Climate Change Scenarios in the Sundarbans, Bangladesh(2018-05-22)Ground-based readings of temperature and rainfall, satellite imagery, aerial photographs, ground verification data and Digital Elevation Model (DEM) were used in this study. Ground-based meteorological information was obtained from Bangladesh Meteorological Department (BMD) for the period 1977 to 2015 and was used to determine the trends of rainfall and temperature in this thesis. Satellite images obtained from the US Geological Survey (USGS) Center for Earth Resources Observation and Science (EROS) website (www.glovis.usgs.gov) in four time periods were analysed to assess the dynamics of mangrove population at species level. Remote sensing techniques, as a solution to lack of spatial data at a relevant scale and difficulty in accessing the mangroves for field survey and also as an alternative to the traditional methods were used in monitoring of the changes in mangrove species composition, . To identify mangrove forests, a number of satellite sensors have been used, including Landsat TM/ETM/OLI, SPOT, CBERS, SIR, ASTER, and IKONOS and Quick Bird. The use of conventional medium-resolution remote sensor data (e.g., Landsat TM, ASTER, SPOT) in the identification of different mangrove species remains a challenging task. In many developing countries, the high cost of acquiring high- resolution satellite imagery excludes its routine use. The free availability of archived images enables the development of useful techniques in its use and therefor Landsat imagery were used in this study for mangrove species classification. Satellite imagery used in this study includes: Landsat Multispectral Scanner (MSS) of 57 m resolution acquired on 1st February 1977, Landsat Thematic Mapper (TM) of 28.5 m resolution acquired on 5th February 1989, Landsat Enhanced Thematic Mapper (ETM+) of 28.5 m resolution acquired on 28th February 2000 and Landsat Operational Land Imager (OLI) of 30 m resolution acquired on 4th February 2015. To study tidal channel dynamics of the study area, aerial photographs from 1974 and 2011, and a satellite image from 2017 were used. Satellite images from 1974 with good spatial resolution of the area were not available, and therefore aerial photographs of comparatively high and fine resolution were considered adequate to obtain information on tidal channel dynamics. Although high-resolution satellite imagery was available for 2011, aerial photographs were used for this study due to their effectiveness in terms of cost and also ease of comparison with the 1974 photographs. The aerial photographs were sourced from the Survey of Bangladesh (SOB). The Sentinel-2 satellite image from 2017 was downloaded from the European Space Agency (ESA) website (https://scihub.copernicus.eu/). In this research, elevation data acts as the main parameter in the determination of the sea level rise (SLR) impacts on the spatial distribution of the future mangrove species of the Bangladesh Sundarbans. High resolution elevation data is essential for this kind of research where every centimeter counts due to the low-lying characteristics of the study area. The high resolution (less than 1m vertical error) DEM data used in this study was obtained from Water Resources Planning Organization (WRPO), Bangladesh. Conference PublicationPublication Reinforced Behavioral Variability and Sequence Learning Across Species(Association for Behavior Analysis International (ABAI), 2012)Previous research shows that reinforcement of variable responding will facilitate sequence learning in rats (Neuringer, Deiss & Olson, 2000) but may interfere with sequence learning in humans (Maes & van der Goot, 2006). The present study aimed to replicate and extend previous research by assessing the role of behavioral variability in the learning of difficult target sequences across 3 species: humans (n = 60), hens (n = 18) and possums (n = 6). Participants were randomly allocated to one of three experimental conditions (Control, Variable, Any). In the Control conditions sequences were only reinforced if they were the target sequence, in the Variability conditions sequences were concurrently reinforced on a Variable Interval 60-s schedule if the just entered sequence met a variability criterion, and in the Any condition sequences were concurrently reinforced on a Variable Interval 60-s schedule for any sequence entered. The results support previous findings with animals and humans; hens and possums were more likely to learn the target sequence in the Variability condition, and human participants were more likely to learn the target sequence in the Control condition. The data has been divided into four folders that are aligned with the data chapters for the thesis. These being: (Ch 2) waterhole mapping, (Ch 3) floodplain gullies, (Ch 4) sediment transport and (Ch 5) fish.
A README file is provided for each chapter which contains a description of the individual datasets and a list of files that make up each dataset.
The data in this archive is a combination of data obtained from desktop studies as well as field work on the Darling River (i.e., the fish data).
Further, fish data were collected on the Darling River between Bourke and Wilcannia. Waterhole mapping was undertaken on the Barwon-Darling between Walgett and Wilcannia. Gully mapping was undertaken on the Barwon-Darling River between Mungindi and Wilcannia. Teachers need to empower students with the means to critically analyse the world around them in order to develop into critical independent thinkers. Students need to be proficient in utilising skills associated with higher levels of thinking, that will empower them with the ability to identify, analyse and evaluate the infinite volume of information available through our rapidly changing digital world. Just as teachers need to take responsibility for the various methods of teaching and instruction in the classroom, it is essential for students to take ownership of the learning process, to ensure future success in university environments, where sustained personal effort and metacognitive skills are fundamental to academic success.
